Shouldn't have to, no. Your Droid recognizes and uses it out-of-the-box. I've also taken my SD card out of the phone to put it in a usb microSD reader and my computer reads it just fine too. No formatting required.
Not out of the box. cyanogen has found a way to get APPS2SD to work on the Droid. You need to be rooted and you may even need to be using his ROM. Check the Hacks section for more info.
