From time to time I see posts about people that can't get a computer to recognize the phone via USB. Personally, I hate having to connect via USB and do it wirelessly. I've been using Sugarsync, and it works perfectly.

Sugarsync allows you to share folders among computers and mobile devices. Because phone storage is minimal, you can specify specifically what files to sync. Files are encrypted while in transit and in storage.

If your rooted, samba fileshare works great to share files within your network.

I've got my sdcard and sdcard-ext as mapped drives.

Put dropbox on both. Put the file you want to transfer into a dropbox folder on your Pc. Open dropbox on your droid, select the file and it downloads to your droid. Use something like astro to move the file if needed.
Dropbox and astro are both free.
